FGF8/17/18 cis-regulatory region drives a dominant negative form of Suppressor of Hairless, a transcription factor known to mediate Notch signalling. Ci-Su(H)DBM was generated by introducing mutations R218E, R220E, R227E and Y228S into the DNA-binding domain of Ci-Su(H) by overlap- extension PCR. Mutations at these four sites result in a DNA-binding mutant that acts in a dominant-negative manner (Hudson and Yasuo 2006).
